Matrixyl (palmitoyl pentapeptide-4, Pal-KTTKS) consists of the pentapeptide sequence Lys-Thr-Thr-Lys-Ser conjugated to a palmitic acid chain for enhanced skin penetration. The KTTKS sequence is a fragment of the C-terminal propeptide of type I procollagen — when collagen is broken down, this fragment acts as a matrikine (ECM-derived signaling molecule) that triggers the wound-repair cascade. It signals dermal fibroblasts via integrin receptors (particularly αvβ5) to upregulate synthesis of collagen types I, III, and IV, fibronectin, elastin, and glycosaminoglycans. Clinical trials demonstrate significant improvement in wrinkle depth and skin roughness after 4-12 weeks of twice-daily application.
